Hi,
I am currently in the USA on a B-2 visitor visa. I have extended my I-94 once. I would like to know can i extend it again.

Yes you can apply to have it extended. It may not be approved though without sufficient justification for another extension.

Sister filed citizenship for me but it is taking too long to get it. My son is now old enough to file for me. Can he do it now?

If your son is 21 and a US citizen he can file for you now. This will have no adverse effect on the other petition that was filed.

I recommend filing the I-130 for your wife soon. Once you become a US citizen the petition will automatically convert.
