Your can refuse, but your employer can require it as a term of employment, and you can be fired. Your call I suppose.

Is a private business and they can set their own terms. They may owe you back money if you prepaid, but they can set their own standards.

Assuming the leave is for medical purposes, the employer can indeed require you to use it whether or not it is in the handbook.
