Hello, I had a form I 485 that was denied in March 2012. Can I file another I 485 based on marriage with us citizen?

Does your DS-2019 say that you are subject to the 2-yr foreign residency requirement? Does the visa in your passport say that you are?

No. The requirements for naturalization are different. You must wait 5 or 3 years from when you first became a green card holder.

No. You're eligible to apply for an EAD but are NOT authorized to work without an EAD card.
