You can only complete this once you are "in the process of returning to the US" but you can start whenever you like. The initial petition takes about 6 months to be approved, and then the second "consular" phase can be as fast or slow as you want. Or rather as slow or snail-like as you want. Fast would be an exaggeration.
You can file an I-130 visa petition for her. After it is approved you can then apply for consular processing. Once that is complete your wife can enter the US as a permanent resident. Once she has been a permanent resident for three years she can become a US citizen by filing an N-400.

You need to complete your sponsorship of her permanent residence before returning to the US. THis process will take about 8 - 12 months, so you should plan accordingly. YOu will also have to prove that your domicile (permanent place of residence) is in the USA--this can be tricky if you have been in Canada and obtained citizenship there.
