Save the e-mail as it might be evidence of some other type of claim but not likely defamation. Rude or belittling are not defamatory, patently false fact and damaging your reputation to others is the foundation of defamation.
Not likely as defamation requires a false statement of fact published to a 3rd party which causes you damages.
Sounds like perhaps an age discrimination case or other. Hope you sent a copy to yourself. It's less defamation than a workplace employment condition claim--consult an attorney who handles employment discrimination issues.
